LIPSHUTZ,~ILLIAM, AND SIDNEY COHEN. Interaction of gastrin I and secretin on gastrointestinal circular muscle. Am. J. Physiol. 222(3) : 775-781. 1972.-The effect of gastrin I and secretin was studied on the in vitro circular smooth muscle of the lower esophageal sphincter (LES), antrum, and pylorus of the opossum. Each muscle was studied at its length of optimal tension development, L,, as determined by length-tension diagrams. At Lo, full-dose response curves were constructed for gastrin I and secretin, alone and in combination. Gastrin I stimulated LES and antral muscle, and secretin produced a selective competitive type of antagonism to this gastrin I response. Pyloric circular muscle did not respond to gastrin I. Secretin, over a wide dose range, had little effect on antral and LES muscle. PyIoric circular muscle was stimulated by secretin, and gastrin I produced a selective, noncompetitive type of antagonism to this secretin response. We conclude that the action and interaction of gastrin and secretin on upper gastrointestinal circular smooth muscle differ at each region. It is suggested that these hormonal effects that have been demonstrated in vitro determine, in part, the motor function of these separate anatomic regions in vivo.

The hormonal regulation of pyloric sphincter function.

The purpose of this study was to evaluate the hormonal control of pyloric sphincter function. Studies were performed on both pyloric circular muscle, in vitro, and the human pylorus, in vivo. Full dose-response curves to gastrin I, cholecystokinin, and secretin were constructed for the pyloric muscle of the opossum studied at its length of optimal tension development, Lo. Both cholecystokinin a...

Gastrin inhibits secretin-induced ductal secretion by interaction with specific receptors on rat cholangiocytes.

We assessed the effect of gastrin on ductal secretion in normal and bile duct-ligated (BDL) rats. The effect of gastrin on ductal secretion was examined in the presence of proglumide, a specific antagonist for gastrin receptor (GR). We isolated pure cholangiocytes from normal and BDL rats and assessed gastrin effects on secretin receptor (SR) gene expression and intracellular adenosine 3',5'-cy...

Hormonal regulation of human lower esophageal sphincter competence: interaction of gastrin and secretin.

The interaction of gastrin and secretin, in the regulation of human lower esophageal sphincter competence, was studied in 54 normal subjects. A dose-response curve, for the lower esophageal sphincter, was constructed from the rapid intravenous injections of synthetic gastrin I (amino acid sequence 2-17). This curve was sigmoid shaped and showed a peak response that was 460.0 +/-24.0% (mean +/-2...

Inhibitory Effect of Secretin and Cholecystokinin on Heidenhain Pouch Responses to Gastrin Extract and Histamine.

Rapid intravenous injections of secretin and cholecystokinin (Vitrum) inhibited the acid response of Heidenhain pouch dogs to continuous intravenous gastrin extract. Cholecystokinin caused greater inhibition than secretin. Responses to small doses of histamine were inhibited by cholecystokinin but not by secretin.

Role of GRPergic neurons in secretin-evoked exocrine secretion in isolated rat pancreas.

Effects of intrapancreatic gastrin-releasing peptide (GRP)-containing neurons on secretin-induced pancreatic secretion were investigated in the totally isolated perfused rat pancreas. Electrical field stimulation (EFS) increased secretin (12 pM)-induced pancreatic secretions of fluid and amylase. EFS induced a twofold increase in GRP concentration in portal effluent, which was completely inhibi...
